Strain Description

：

1. A modified Il15ra transgene was constructed by replacing the IL-15Ra IC domain with the TCRa IC domain in mouse bacterial artificial chromosome (BAC) clone (RP24-148D16). TCRa IC domain composed of TCRa IC domain, FLAG, a stop codon, and carrying an additional A nucleotide in exon 6 at a site immediately 3' to the coding sequence of the transmembrane domain. The ∼54-kb BAC fragment containing the modified Il15ra was microinjected into the KOzygotes.
2. In the case of resting Tg1 BMDCs, the level of IL-15Ra on the cell surface or in cell lysate was below detection.
3. The level of surface IL-15Ra follows the order of WT>Tg6 (RMRC13291) >Tg1 (this strain)>KO. The same order was observed for IL-15Ra on splenic CD11b⁺ DCs, F4/80⁺CD11b⁺macrophages as well as on BMDMs and stromal cells.
